Evolving enzymes for nonnatural amino acids

A new study by the Söll lab in cooperation with David Liu’s lab at Harvard reveals the structure of an elusive functional domain of pyrrolysyl-tRNA synthetase, an enzyme that is widely used for genetic encoding of unnatural amino acids in genome of a live cell. The authors of the study also have established a method of continuous directed evolution of aminoacyl-tRNA synthetases to improve activity and specificity of aminoacyl-tRNA synthetases to unnatural amino acids. The study was published in two side-by-side papers in Nature Chemical Biology.
